Cisapride

Cisapride is a gastroprokinetic agent and third-generation benzamide that is used as a motility-enhancement drug, to help increase motility in the upper gastrointestinal tract. Cisapride is a serotonergic agonist which activates acetylcholine release from the mesenteric plexus, which in turn stimulates GI smooth-muscle.

Caution: Cisapride should not be used when increased GI motility may be harmful (mechanical obstruction, impaction, potential perforation or GI hemorrhage).

Caution: Cisapride should not be used with ketoconazole, itraconazole, IV miconazole, or troleandomycin, erythromycin, fluconazole and clarithromycin. These combinations may result in potentially fatal ventricular arrhythmias.

Caution: If used with cimetidine, it may increase the absorption of cisapride.
  • 0.5-1.5 mg/kg given orally, every 8 hours.
  • Can be harmful when mechanical obstruction, impaction, or potential perforation or GI hemorrhage is the cause of GI tract motility.
